Background Information: The Killer Cell Lectin Like Receptor C1 (KLRC1, also known as CD159a) is a member of the NKG2 family, a group of transmembrane proteins preferentially expressed in natural killer (NK) cells. KLRC1 forms a complex with KLRD1 (CD94), and is implicated in the reccognition of the MHC class I HLA-E molecules in NK cells. KLRC1 enables cytotoxic cells to monitor the expression of MHC class I molecules in healthy cells and to tolerate self, as well as monitoring the activation and effector functions of NK cells. In viral infections and some tumor microenvironments, KLRC1 is a key target to downregulate NK cell cytotoxicity and allow tumor progression and viral escape from immune surveillance.
