Endotoxin Level: <1.0 EU per μg of the protein as determined by the LAL method

Activity (Specifications/test method): N/A

Biological activity: Activity of CD16a was measured by its binding ability in a functional ELISA. Immobilized human CD16a at a concentration of 10 μg/ml (100 μl/well) can bind biotinylated human IgG1.

Results: N/A

Binding Capacity: N/A

Unit Definition: N/A

Molecular Weight: The recombinant human CD16a consists of 203 amino acids and has a predicted molecular mass of 23.3 kDa. The protein migrates as a band at ~45.2 kDa due to glycosylation, in SDS-PAGE under reducing conditions.

Concentration: N/A

Appearance: Lyophilized protein

Physical form description: Lyophilized from sterile PBS, pH 7.4, 5% trehalose, 5% mannitol and 0.01% Tween-80.

Reconstitution Instructions: Reconstitute in sterile water to a concentration of 250 µg/ml by adding 200 µl water to 50 µg of protein.

Background Information: CD16a/FCGR3A is a low affinity receptor for the Fc region of IgG. The two genes for human FCGR3A encode for CD16A and CD16B. CD16a is a transmembrane protein found on the surface of natural killer cells, activated monocytes and macrophages and a subset of T cells. CD16a is involved in phagocytosis, secretion of enzymes and inflammatory mediators, antibody dependent cytotoxicity and clearance of immune complexes. Aberrant expression or mutations of CD16a can cause susceptibility to recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.

Amino acid sequence: The DNA sequence encoding the extracellular domain (Met 1-Gln 208) of human CD16a (P08637-1) precursor was expressed with a C-terminal polyhistidine tag. The predicted N terminal is Gly 17.

Handling: Centrifuge the vial prior to opening.

Usage: For Research Use Only! Not to be used in humans
