Background Information: Trophoblast glycoprotein, also known as TPBG or 5T4, is a human cell surface protein encoded by a TPBG gene and an antagonist of the of Wnt/β-catenin signaling pathway. TPBG/5T4 is rarely expressed in normal adult tissues, but is present at high levels in placenta and in most common tumors, typically more than 80% of carcinomas of the kidney, breast, colon, prostate, and ovary. Due to its selective expression, it is often used as a prognostic aid in cancer cases. Recently, it has been shown that TPBG inhibits Wnt/β-catenin signaling, a signaling system central to many developmental and pathological processes and therefore serves as a therapeutic target of several anticancer agents currently in clinical development.
